Wellness Peptides Are Trending: Are They Safe?

In gyms, longevity forums, and on social media, peptides have long been promoted as miracle cures for faster recovery, fat loss, better sleep, and so-called “antiaging.” Suppliers often promise dramatic results with minimal risk despite limited or no scientific evidence.

Many of these substances are neither approved nor adequately studied in humans. Numerous products also come from questionable sources, making their composition uncertain and leaving their potential adverse effects poorly understood.

Diabetes Drugs

The hype surrounding injectable peptides has several drivers. One is the success of GLP-1 receptor agonists such as semaglutide and tirzepatide. These drugs have shown that endogenous signaling molecules and their synthetic derivatives can have powerful effects on appetite, weight, and metabolism. This has drawn attention to a broader class of compounds: peptides, which are short chains of amino acids that often act as signaling molecules in the body.

The biohacking market capitalizes on this interest. Many individuals extrapolate the established effects of approved peptides or peptide analogs to an entire class of unapproved substances. The reasoning is straightforward: If GLP-1 receptor agonists can have powerful effects on weight and metabolism, perhaps other peptides could also improve recovery, slow aging, or enhance performance.

These substances include BPC-157, a synthetic peptide promoted for regenerative effects; TB-500, a synthetic variant of a peptide derived from thymosin beta 4; CJC-1295, a growth hormone-stimulating peptide; ipamorelin, a growth hormone secretagogue; epitalon, a synthetic tetrapeptide promoted for antiaging effects; GHK-Cu, a copper-binding peptide associated with skin and tissue regeneration; Selank and Semax, neuroactive peptides promoted for anxiolytic and cognitive effects, respectively; Melanotan II, a synthetic analog of melanocyte-stimulating hormone; and retatrutide, an experimental multi-receptor agonist being investigated as part of the newer class of incretin-based therapies.

BPC-157 and TB-500 are promoted as treatments that can repair tissue, heal tendons, and reduce inflammation. CJC-1295 and ipamorelin are marketed as natural growth hormone boosters, while Melanotan II is promoted not only for tanning but also for increasing libido. Epitalon has been linked to telomeres and purported antiaging effects. GHK-Cu is promoted for skin, hair, and nail health and is also claimed to accelerate wound healing. Retatrutide, by contrast, has pharmacologic properties similar to those of newer incretin therapies and is being clinically investigated for obesity and metabolic disorders.

However, in the rapidly expanding peptide market, biological plausibility is often quickly converted into therapeutic claims. Many purported benefits are based primarily on cell culture experiments, animal studies, small early-stage trials, or anecdotal reports on social media. In many cases, reliable evidence on safety and efficacy is lacking, particularly for self-administration by healthy individuals. The uncertainty is even greater when multiple substances are combined and injected without medical supervision. A reliable assessment of the benefits and risks is usually not possible in these circumstances.

Limited Evidence

The discrepancy is particularly clear with BPC-157. PubMed lists studies and reviews examining its potential regenerative effects. A 2025 pilot study found that intravenous administration was tolerated in the short term by only two healthy adults. Although this finding is of interest, it provides no evidence of efficacy and does not support widespread use. A review therefore concluded that BPC-157 should be considered experimental until well-designed clinical trials are conducted.

The situation is similar for many other “longevity” peptides. Early clinical data on CJC-1295 and ipamorelin have examined effects on growth hormone pathways, but there is no robust evidence that healthy individuals can safely improve recovery, body composition, or aging-related processes over the long term. The FDA has noted limited clinical data, a lack of safety information, and potential risks for immune reactions or impurities with several of these substances.

Melanotan II illustrates the potential risks in this poorly regulated area. PubMed contains case reports of systemic toxicity involving sympathomimetic symptoms, rhabdomyolysis, and renal dysfunction, as well as reports of priapism, renal infarction, and melanoma occurring in temporal association with its use.

The concern is not limited to adverse effects from the peptide itself. Uncertain product quality is another major concern. Individuals who order “research peptides” online may not know whether a product contains the stated substance, whether the dose is accurate, whether it contains impurities, or whether it is sterile. This is particularly concerning with the injectable products because microbial contamination, incorrect concentrations, or foreign substances can be introduced directly into the body.

Another concern is peptide “stacking,” in which individuals combine multiple substances. This makes adverse effects and drug interactions even harder to predict. The effects of combining growth hormone secretagogues, melanocortin analogs, GLP-1 receptor agonists, and so-called regenerative peptides are not well understood.

Self-Experimentation

Peptides are not inherently unproven therapies. Some widely used medications are peptides or peptide analogs, including insulin, GLP-1 receptor agonists, and certain hormone therapies. The difference lies in their development, regulatory approval, quality control, established indications, and medical supervision.

The current wellness trend often reverses this process: Influencers make the promise first, followed by self-administration and, perhaps later, attempts to establish the evidence. That is where the concern lies. For many popular peptides, reliable human data, validated dosing, long-term safety information, and regulatory approval are still lacking.
